Armillaria root rot (ARR) poses a significant threat to the long-term productivity of stone-fruit and nut crops in the predominant production area of the United States. To mitigate this issue, the development of ARR-resistant and horticulturally-acceptable rootstocks is a crucial step towards the maintenance of production sustainability. To date, genetic resistance to ARR has been found in exotic plum germplasm and a peach/plum hybrid rootstock, ’MP-29‘. However, the widely-used peach rootstock Guardian® is susceptible to the pathogen. To understand the molecular defense mechanisms involved in ARR resistance in Prunus rootstocks, transcriptomic analyses of one susceptible and two resistant Prunus spp. were performed using two causal agents of ARR, including Armillaria mellea and Desarmillaria tabescens. The results of in vitro co-culture experiments revealed that the two resistant genotypes showed different temporal response dynamics and fungus-specific responses, as seen in the genetic response. Gene expression analysis over time indicated an enrichment of defense-related ontologies, including glucosyltransferase activity, monooxygenase activity, glutathione transferase activity, and peroxidase activity. Differential gene expression and co-expression network analysis highlighted key hub genes involved in the sensing and enzymatic degradation of chitin, GSTs, oxidoreductases, transcription factors, and biochemical pathways likely involved in Armillaria resistance. These data provide valuable resources for the improvement of ARR resistance in Prunus rootstocks through breeding.

In this review, major peach diseases from China, Spain, and USA are described in detail for the benefit of producers, consultants, researchers, and other interested parties. Minor diseases of concern in these countries are also described. Current progress on pathogen resistance to major chemical classes of fungicides as well as current resistance management practices are discussed. Specific cultural practices applied in China, Spain, and USA are also described to provide an overview of peach disease management. A ‘Future Outlook’ section is included at the end of this review to highlight the challenges and opportunities for disease management in the future.

Trichoderma is a genus of filamentous fungi with several species widely studied and used as biological control agents (BCAs) in agriculture. With respect to fungal disease control, Trichoderma spp. can use different mechanisms of action, including mycoparasitism, antibiosis, competition and/or activation of plant defences. Armillaria spp. are the causal pathogens of Armillaria root rot (ARR) disease, which is widely distributed and of great economic and environmental importance in agriculture and forestry. ARR can be devastating to plant health, through colonisation and rotting of the root system and collar of the infected plant/tree, which can cause a reduction in yield and eventually death. Armillaria can live as a saprophyte that survives on dead plant material, making eradication of ARR extremely difficult. In an exhaustive analysis of work published to date on the control of Armillaria spp. by Trichoderma spp., using the WoS and Scopus databases, we identified 31 papers. We discuss these reports in light of their key findings, including number of publications per year, the countries where they were published, the journals and the citations obtained so far. Although the vast majority do not identify the mechanism of action involved in control, some describe mycoparasitism, antibiosis, competition or modification of the rhizospheric microbiota as being responsible for effective biological control of the pathogen. Further research into these mechanisms and additional in-depth analysis of less studied or currently unidentified mechanisms, such as the activation of plant defences, would be highly beneficial to our understanding of Trichoderma as a BCA of Armillaria. Species of Armillaria are distributed globally and include some of the most important pathogens of forest and ornamental trees. Some of them form large long-living clones that are considered as one of the largest organisms on earth and are capable of long-range spore-mediated transfer as well as vegetative spread by drought-resistant hyphal cords called rhizomorphs. However, the virus community infecting these species has remained unknown. In this study we used dsRNA screening and high-throughput sequencing to search for possible virus infections in a collection of Armillaria isolates representing three different species: Armillaria mellea from South Africa, A. borealis from Finland and Russia (Siberia) and A. cepistipes from Finland. Our analysis revealed the presence of both negative-sense RNA viruses and positive-sense RNA viruses, while no dsRNA viruses were detected. The viruses included putative new members of virus families Mymonaviridae, Botourmiaviridae and Virgaviridae and members of a recently discovered virus group tentatively named "ambiviruses" with ambisense bicistronic genomic organization. We demonstrated that Armillaria isolates can be cured of viruses by thermal treatment, which enables the examination of virus effects on host growth and phenotype using isogenic virus-infected and virus-free strains. Avocado has great socioeconomic importance in Mexico because of the benefits it generates for the production chain participants and the significant foreign exchange earnings engendered by the export of its fruit. However, this crop has phytosanitary problems, caused mainly by fungi, among which the genus Fusarium stands out. The objective of this study was to characterize Fusarium species that caused root rot in avocado trees in Michoacan, Mexico. In 19 isolates of Fusarium spp., polymerase chain reactions (PCR) with primers coding for elongation factor and calmodulin genes were performed. These sequences were compared in homology using BLAST analysis and aligned in MEGA 6.0. Cladograms were created based on maximum verisimilitude. The pathogenicity of the isolates was evaluated based on their virulence and severity in the avocado plants. Morphological and molecular analyses showed that 15 isolates belonged to F. oxysporum Schl and four to F. solani Mart. All isolates were pathogenic, with virulence ranging from 16 to 56 days. All isolates produced root rot and yellowing of leaves, with 63% producing wilting and 16% producing apical necrosis, the latter being the most severe. [ABSTRACT FROM AUTHOR]

The primary focus of the stone fruit rootstock program at Byron, GA, has been the development of disease-resistant rootstocks for peach (Prunus persica L. Batsch). Historically peach tree short life (PTSL), aka bacterial canker complex, and Armillaria root rot (ARR) have been the two most important causes of premature mortality of commercial peach trees in the southeastern United States. Guardian®, a seedling peach rootstock, was cooperatively released in 1993 by the U.S. Department of Agriculture (USDA)-Agricultural Research Service (ARS) and Clemson University. It has since been widely adopted by the southeastern peach industry. As a result, trees losses to PTSL have declined sharply. However, Guardian, like most other peach seedling rootstocks, is susceptible to ARR. ARR has now moved to the forefront as the primary cause of premature peach tree death in the Southeast. In response to this threat, the USDA-ARS in cooperation with the University of Florida, released 'Sharpe', a plum hybrid rootstock in 2007. Despite its broad disease resistance, 'Sharpe' proved unsuited for widespread commercial utilization due to its relatively poor cropping performance. In 2011, 'MP-29', a semidwarf, clonal, plum × peach hybrid, was released for commercial trial. 'MP-29's broad disease and nematode resistance, in combination with its dwarfing ability and excellent productivity, offered great promise for use in this production area and in others suffering from similar issues. Western redcedar (WRC) trees of 48-49 years in age grown in monoculture and in admixtures with birch, Douglas-fir and pine in the interior of British Columbia were sampled for wood density and the presence and level of damage from butt decay initiated by Armillaria ostoyae in stumped and unstumped plots. Stump removal reduced butt decay incidence in WRC from 18% to 2% and damage from 3.5% of the diameter at base to 0.3% compared to when stumps remained. External lesions on tree bases caused by Armillaria root disease increased the probability of WRC stems having butt decay by 1.25 times for an average-sized lesion. The incidence of butt decay and the level of internal damage to WRC in sampled plots was not related to admixture, tree size or site index. Stump removal controlled the inoculum of Armillaria root disease, thereby reducing incidence and size of WRC basal lesions that probably act as infection courts for other fungi causing butt decay. WRC trees grown in monoculture or with birch had the highest wood densities of all planting mixes tested. Wood density in WRC was lowest at the outer edges of individual WRC trees, in larger trees, and in plots where stumps had not been removed. The lower density WRC wood is thought to occur by overtopping from fast-growing species in admixture and depletion of soil moisture affecting latewood formation. Prunus dulcis (almond) is one of the most susceptible horticultural crops to Armillaria root disease. Resistance to Armillaria mellea and Armillaria tabescens, the geographically isolated causal fungi that attack almond and closely related Prunus persica (peach), has been evaluated in studies of almond, peach, and other Prunus rootstocks, but not in one comprehensive study. We evaluated the relative resistance to A. mellea and A. tabescens of six clonally propagated almond and peach rootstocks (Bright's 5, Empyrean 1, Hansen 536, Krymsk 1, Krymsk 86, and Lovell) in comparison with that of clonally propagated Marianna 2624 rootstock (resistant control) and clonally propagated Nemaguard rootstock (susceptible control). Replicate clones used in the growth chamber assay were micropropagated and rooted in vitro before inoculating the culture medium with Armillaria spp. At 2 months, the most resistant and susceptible rootstocks were Krymsk 86 and Hansen 536, respectively, with 27% vs. 89% mortality. This finding was consistent among two isolates of A. mellea and one isolate of A. tabescens in three replicate experiments. Our finding of low mortality among Krymsk 86, Krymsk 1, and Marianna 2624, which all share Prunus cerasifera (Myrobalan plum) parentage, is consistent with past reports of resistance in the field to A. mellea, but conflicts with reports of susceptibility to A. tabescens. Resistance to A. tabescens of genotypes with Myrobalan plum parentage in our assay may reflect the simplified rooting environment of tissue culture medium, which does not perfectly mimic a field trial, in which biotic and abiotic factors may affect host resistance. Armillaria root rot, one of the most dangerous diseases in our forests, is caused in Poland mainly by Armillaria ostoyae, especially severe in young Scots pine stands, established after broadleaved stands or with participation of broadleaved species. In Forest District Zielonka young stands are severly affected by Armillaria root rot. Only one species, A.ostoyae, was found in the young (8-14 yrs) Scots pine stands, despite the presence of other Armillaria species in the district. The pathogen's frequent occurrence may be due, inter alia, to favouring environmental factors.

Background: Armillaria is a globally distributed mushroom-forming genus composed primarily of plant pathogens. Species in this genus are prolific producers of rhizomorphs, or vegetative structures, which, when found, are often associated with infection. Because of their importance as plant pathogens, understanding the evolutionary origins of this genus and how it gained a worldwide distribution is of interest. The first gasteroid fungus with close affinities to Armillaria--Guyanagaster necrorhizus--was described from the Neotropical rainforests of Guyana. In this study, we conducted phylogenetic analyses to fully resolve the relationship of G. necrorhizus with Armillaria. Data sets containing Guyanagaster from two collecting localities, along with a global sampling of 21 Armillaria species--including newly collected specimens from Guyana and Africa--at six loci (28S, EF1α, RPB2, TUB, actin-1 and gpd) were used. Three loci--28S, EF1α and RPB2--were analyzed in a partitioned nucleotide data set to infer divergence dates and ancestral range estimations for well-supported, monophyletic lineages. Results: The six-locus phylogenetic analysis resolves Guyanagaster as the earliest diverging lineage in the armillarioid clade. The next lineage to diverge is that composed of species in Armillaria subgenus Desarmillaria. This subgenus is elevated to genus level to accommodate the exannulate mushroom-forming armillarioid species. The final lineage to diverge is that composed of annulate mushroom-forming armillarioid species, in what is now Armillaria sensu stricto. The molecular clock analysis and ancestral range estimation suggest the most recent common ancestor to the armillarioid lineage arose 51 million years ago in Eurasia. A new species, Guyanagaster lucianii sp. nov. from Guyana, is described. Conclusions: The armillarioid lineage evolved in Eurasia during the height of tropical rainforest expansion about 51 million years ago, a time marked by a warm and wet global climate. Species of Guyanagaster and Desarmillaria represent extant taxa of these early diverging lineages. Desarmillaria represents an armillarioid lineage that was likely much more widespread in the past. Guyanagaster likely evolved from a gilled mushroom ancestor and could represent a highly specialized endemic in the Guiana Shield. Armillaria species represent those that evolved after the shift in climate from warm and tropical to cool and arid during the late Eocene. No species in either Desarmillaria or Guyanagaster are known to produce melanized rhizomorphs in nature, whereas almost all Armillaria species are known to produce them. The production of rhizomorphs is an adaptation to harsh environments, and could be a driver of diversification in Armillaria by conferring a competitive advantage to the species that produce them. Brassicaceous plants rich in glucosinolates have been used as biofumigants for the management of soilborne pathogens. Efficacy of Brassica plant tissue has mainly been attributed to toxic isothiocyanates released upon the hydrolysis of glucosinolates. Management of Phytophthora cinnamomi, the causal agent of oak root rot in rangeland ecosystems using biofumigation, is promising, but requires further validation. The biofumigation activity of 14 brassicaceous plants was evaluated under experimental conditions. All evaluated plants rich in sinigrin suppressed (100%) the mycelial growth of P. cinnamomi, while plants rich in aromatic or other aliphatic glucosinolates had little or no suppressive effect. Simulating soil amendment in field conditions, the effects on natural soil artificially infested with P. cinnamomi chlamydospores were examined with Brassica juncea, Eruca vesicaria and Lepidium sativum, three species with different glucosinolate profiles. Only B. juncea decreased the viability of chlamydospores significantly in comparison with untreated soil only 1 day after biofumigation, whereas E. vesicaria needed 8 days to reach significance and L. sativum had no effect at all. Despite the decreases in soil inoculum, biofumigation with B. juncea did not prevent the root infections in a highly susceptible host ( Lupinus luteus). However, biofumigation with plants rich in sinigrin, such as B. juncea, decreased P. cinnamomi soil inoculum under the experimental minimum threshold for oak disease expression. Although biofumigation should be considered as an effective measure to be incorporated in integrated control of the oak disease, biofumigation by itself would not be effective enough for the substantial suppression of P. cinnamomi inoculum. During May 2010, sporocarps of what appeared to be an Armillaria sp. were found in large clumps in historic Kirstenbosch Botanical Gardens on the foot of Table Mountain, Cape Town, South Africa. These sporocarps could be physically linked to the roots of unidentified dead trees and Protea spp. The aim of this study was to identify the Armillaria sp. found fruiting in Kirstenbosch. To achieve this goal isolates were made from the mycelium under the bark of dead roots linked to sporocarps. The ITS and IGS-1 regions were sequenced and compared to sequences of Armillaria spp. available on GenBank. Cladograms were generated using ITS sequences to determine the phylogenetic relationship of the isolates with other Armillaria spp. Sequence comparisons and phylogenetic analyses showed that the isolates represented A. mellea. They were also identical to isolates of this species previously discovered in the Company Gardens in South Africa and introduced from Europe apparently by the early Dutch Settlers. Armillaria mellea is alien and apparently invasive in Cape Town, fruits profusely and has the potential to spread to sensitive native forests on the foothills of the City.

Armillaria root disease is a significant forest health concern in the Atlantic forest ecosystems in Spain. The damage occurs in conifers and hardwoods, causing especially high mortality in young trees in both native forests and plantations. In the present study, the distribution of Armillaria root disease in the forests and plantations of the Basque Country is reported. Armillaria spp. were more frequently isolated from stands with slopes of 20–30% and west orientation, acid soils with high permeability, deciduous hosts, and a rainfall average above 1800 mm. In a large-scale survey, 35% of the stands presented Armillaria structures and showed disease symptoms. Of the isolated Armillaria samples, 60% were identified using molecular methods as A. ostoyae, 24% as A. mellea, 14% as A. gallica, 1% as A. tabescens, and 1% as A. cepistipes. In a small scale sampling, population diversity was defined by somatic compatibility tests and Universally Primed-PCR technique. Finally, the pathogenicity of A. mellea, the species with the broadest host range, was determined on different tree species present in the Atlantic area of Spain in order to determine their resistance levels to Armillaria disease. A significant difference in disease severity was observed among tree species (p < 0.001), with Pinus radiata being the most susceptible tree species and Cryptomeria japonica the most resistant to A. mellea. Two studies were undertaken in a young New Zealand Pinus radiata stand to find a biological control agent for armillaria root disease caused by Armillaria novae-zelandiae. Fresh thinning stumps were inoculated with saprophytic basidiomycete species as wooden dowel cultures or as homogenized aqueous mycelial suspensions, while all stumps were also inoculated with dowel cultures of A. novae-zelandiae. After a period of between one and two years, no differences were found among test fungi in the percentage of Armillaria species isolated from stumps. However, inoculation of Stereum sanguinolentum as homogenate, and of Phlebiopsis gigantea, Sistotrema brinkmannii, Hypholoma acutum and Rigidoporus concrescens as dowel cultures, significantly increased their isolation incidence in comparison with stumps not treated by these species. Phlebiopsis gigantea, Resinicium bicolor and R. concrescens were also isolated with greater frequency nearer to their dowel inoculation points. Results suggest that P. gigantea and S. sanguinolentum should be included in further testing. However, a future trial is likely to be more effective if evaluation is directed towards the portion of the stump below the soil surface, particularly the root system. [ABSTRACT FROM AUTHOR]

This work investigated the activated carbons (ACs) derived from three kinds of biomass waste carbon precursors, i.e.Phragmites australis(PA), mushroom root (MR), and chicken feather (CF) with H4P2O7activation. The effect of different precursors on porous texture, surface chemistry, and adsorption capacities of the derived ACs were comparatively studied. The pore structure and chemical properties of the ACs were characterized by N2desorption/adsorption isotherms, element analysis, FTIR, XPS and Boehm’s titration methods. It was found that the surface area and pore volume of AC-PA (SBET = 986.8 m2/g,Vtot = 1.07 cm3/g) and AC-MR (SBET = 950.1 m2/g,Vtot = 1.02 cm3/g) were about 1.7 times higher than those of AC-CF (SBET = 471.4 m2/g,Vtot = 0.28 cm3/g). AC-PA (2.488 mmol/g) possessed more acidic oxygen functionalities than AC-MR (1.736 mmol/g) and AC-CF (1.858 mmol/g), while AC-CF had most basic functional groups. Batch adsorption studies were performed to compare the adsorptive abilities of different ACs for trimethoprim (TMP). It was found that the adsorption affinities of TMP followed an order of AC-PA (244.9 mg/g) > AC-MR (165.6 mg/g) > AC-CF (124.2 mg/g). [ABSTRACT FROM PUBLISHER]

Ectomycorrhizal symbiosis is essential for the life and health of trees in temperate and boreal forests where it plays a major role in nutrient cycling and in functioning of the forest ecosystem. Trees with ectomycorrhizal root tips are more tolerant to environmental stresses, such as drought, and biotic stresses such as root pathogens. Detailed information on these molecular processes is essential for the understanding of symbiotic tissue development in order to optimize the benefits of this natural phenomenon. Next generation sequencing tools allow the analysis of non model ectomycorrhizal plant-fungal interactions that can contribute to find the “symbiosis toolkits” and better define the role of each partner in the mutualistic interaction. By using 454 pyrosequencing we compared ectomycorrhizal cork oak roots with non-symbiotic roots. From the two cDNA libraries sequenced, over 2 million reads were obtained that generated 19552 cork oak root unique transcripts. A total of 2238 transcripts were found to be differentially expressed when ECM roots were compared with non-symbiotic roots. Identification of up- and down-regulated gens in ectomycorrhizal roots lead to a number of insights into the molecular mechanisms governing this important symbiosis. In cork oak roots, ectomycorrhizal colonization resulted in extensive cell wall remodelling, activation of the secretory pathway, alterations in flavonoid biosynthesis, and expression of genes involved in the recognition of fungal effectors. In addition, we identified genes with putative roles in symbiotic processes such as nutrient exchange with the fungal partner, lateral root formation or root hair decay. These findings provide a global overview of the transcriptome of an ectomycorrhizal host root, and constitute a foundation for future studies on the molecular events controlling this important symbiosis. Armillaria species display diverse ecological roles ranging from beneficial saprobe to virulent pathogen. Armillaria solidipes (formerly A. ostoyae), a causal agent of Armillaria root disease, is a virulent primary pathogen with a broad host range of woody plants across the Northern Hemisphere. This white-rot pathogen grows between trees as rhizomorphs and attacks sapwood as mycelial fans under the bark. Armillaria root disease is responsible for reduced forest productivity due to direct tree mortality and non-lethal infections that impact growth. Here, we characterize a transcriptome of a widespread, virulent genet (vegetative clone) of A. solidipes isolated from a mycelial fan on a natural grand fir ( Abies grandis) sapling in northern Idaho, USA. cDNA from polyA+-purified total RNA was sequenced using a single-end read approach on the Illumina GAIIx platform which generated 24 170 384 reads. A BLASTx search against the NCBI nr database using 39 943 de novo assembled contigs resulted in 24 442 sequences with significant hits (e-value < 1e−3), predominantly to fungi (85%). A filtered data set of 20 882 assembled transcripts that encoded putative homologous fungal proteins was created and used for all subsequent analyses. Signal P identified 10 668 putative signal peptides from these fungal transcripts, and 14 360 were annotated with gene ontology terms. Several sequences showed strong homology to annotated genes with functions in pathogenesis, specifically those involved in plant cell wall degradation and response to the post-infection host environment. This transcriptome contributes to the growing body of resources for studies on fungal pathogens of woody plants, and our results provide useful insights towards identifying specific genes with potential roles associated with pathogenesis and other metabolic functions. Incidence of peach [ Prunus persica (L.) Batsch] tree mortality attributed to Armillaria root disease was assessed from 2009 to 2011 in 15 orchards in the State of Mexico, Mexico. Incidence increased gradually every year of assessment, reaching average values of 9.7, 15.3 and 20.3% tree mortality and 23.2, 24.7 and 28.3% disease-impacted area of the orchards during 2009, 2010 and 2011, respectively. The cultivars 'Nemaguard' and 'Criollo of La Goleta', a local rootstock used in the region, were both susceptible to the disease. To identify species of Armillaria isolated from infected peach trees, two nuclear rDNA regions (partial 5.8S-ITS2- LSU D-domains and partial 3′ LSU-IGS1) and the translation elongation factor-1α ( tef-1α) gene were sequenced and compared with sequences of known Armillaria species. DNA sequence analysis from 49 Armillaria isolates revealed that five isolates (10.2%) were Armillaria mellea and eight isolates (16.3%) were Armillaria gallica. DNA sequences from the remaining 36 isolates (73.5%) showed no close similarity to Armillaria sequences in GenBank, and apparently represent an undescribed Armillaria species. This undescribed species was the most widely distributed in the region of study. Separate phylogenetic analyses of the LSU region ( D1- D3 domains concatenated with the partial 3′ end) and the tef-1α region show that the undescribed species is quite distinct from other Armillaria spp. reported in North America.
